Medium is made from glucose, agar and Vogel-Bonner medium E (50X)


Vogel-Bonner Medium E, 50X (50X VBE)

Citric Acid monohydrate

K2HPO4 (anhyd.)

175 g

Add salts and citric acid to warm water in a 2 L flask on stirring hotplate in order listed. Wait for each to dissolved completely before adding the next. Bring volume up to 1 L, loosely cap and autoclave.

Add agar and magnetic stir bar to DI in 2 L flask and bring to boil on hotplate stirrer. Autoclave. Sterile filter 40% glucose and 50X VBE. Add VBE and glucose to agar solution using sterile technique (in a sterile field) cover flask and stir. Pour plates when soln. is mixed.
